The Hydra are located at 38, 39 in the lake. To get the samples you just use the container when you're afflicted with the poison the hydras use.

Do this quest while you have Jaloot with you. Jaloot's Intensity (https://www.wowhead.com/wotlk/spell=52119) gives a swim speed buff of 100%.

This is a poorly designed quest for pet classes, since the hydra need to be focused on you the whole time, and you cannot collect the sample off of your pet

Yes, Like stated above this quest is 100% much easier to do if you use Jaloot's Intensity! To get Jaloot you must be on one of the following quests.

As some have stated, pets will make it evil.
either they only have 1 shot of the debuff you need on you, or its on a hefty cooldown.

Passive pet, pull the mob, (they will cast it early) use the questpot to seale the debuff in it.
Then nuke it down.
They only do this once, so dont wait for another debuff from em. :) if they used it on your pet, kill it and take a new one.

Pop-culture reference: Lost (TV Series 2004)

The island in the middle of the lake has a hatch - with a sequence of 6 numbers.

The numbers are the same as those from Lost, each incremented by 1.

Hydra is also the name of one of the stations on the 'Lost Island'.

This quest will be a little bit complicated if you are hunter.

Don't mess up and first kill hydras, then kill them again for the samples.
You kill hydras, when they poison you use Sample Container you were given by the Colvin Norrington.

If you are hunter you should dismiss your pet or put it on passive, then when you are poisoned re-call it or press attack.

Note of suggestion for Hunters: Put your Pet on Passive, I couldn't collect the poison because he hit my cat first. Same goes if you have another in your party and they don't have the quest; If they hit the Hydra first, you can't collect the poison for the quest item needed.

Hunters, Warlocks, Unholy DKs:

You cant take the poison off your pet...make sure the hydra is targeting you midway through the fight.

Hunters can use distracting shot.

I was dumb. If you are a skinner, (1) kill (2) loot (3) use Sample Container (4) and "then" skin. :P I had to kill 10 because I failed to read quests.

Not sure if it's just me, but when I click Sample Container while I have the hydra sputum effect , it removes the effect but doesn't give any credit towards the quest. ):

You can only sample poison on yourself.
Hold combat pets back until the Hydra poisons you or else the pet gets poisoned and not you.

Looks like the Hydras only spit in melee distance - not from range. You'll have to take a hit or two.

Colvin Norrington at Dorian's Outpost wants you to take 5 Hydra Sputum Samples and kill 5 Bittertide Hydra.

There's no more caustic material in all of the basin than bittertide hydra sputum. I've got special containers to collect just such a thing for Archmage Pentarus' studies. Now all I need is a brave, um... collector.

You look like a hardy, adventurous <class>. How'd you like to make a bit of money and aid Dalaran's cause at the same time?

All you need to do is take this container with you to Bittertide Lake, to the southwest. Engage the hydra and use the container to sample the sputum they shower you with.
