Sounds like an excuse to me. Never in my experience has Ventrilo used enough system resources to impact how smoothly WoW plays. I get a lot more trouble out of my web browser by comparison. If I'm running a lot of applications on top of the game and it's been a few days since the last time I restarted my computer, Vent will sometimes freeze the game briefly when I connect to a server. But after that the game runs perfectly fine for me with moderate settings and a less than stellar computer.

No, it can be Vent that lags people, but it depends purely on the server. Some connections can't handle the high bit rate which some people use on their server for sound quality/ Better sound quality = larger connection strain. SOmetimes Vent servers just have poor connection and high data loss.
Don't just write it off. JUst because you've never been ona server thats lagged you, or your server doesn't, doesn't mean none will.
And this coming from someone who raid leads, and has been on a vent that had such high packet loss it caused lapses of 10 minutes in vent transmissions, and 2-3 seconds of input delay to the game server.

I think it is more a result of what computer they have, and just how old it is. It may be that their system barely has the stats (or, is under the minimum stats) to play WoW as it is, and that running Vent on top of WoW gets to be a problem.
I've seen a lot of raids, though, where "No vent = No loot". These folk are just SoL, if they still refuse to use Vent, I guess. :-/

Network lag or system lag? Network lag could be due to using a better quality codec in vent and, thusly, you need a better upload speed to transmit your voice and download to receive other peoples'. System lag could be due to RAM, processor, sound card and/or drivers, etc...
Given that WoW uses no more than 15kB/s up and down in big battles, therefore 30kB/s total (and that's being fairly generous), you'd need something along the lines of a 256kb/s itnernet connection to be network-lagged by anything... A 8mb/s line, should you get your full 8mb, you have 1024kB/s (aka 1mB/s) to play with.... Yes, one megaBYTE per second. Granted that you'll never achieve this due to protocal headers/footers and general line quality depreciation, and distance, but it's worth noting that not every has, or can afford, high(er) speed internet!
I wouldn't accuse them of lying to avoid having to use, or even download, Vent but, at the same, I wouldn't discount the laziness of some people...

Do you know where I can find more information on this? I have a 640Kb/s line speed, and in big battles, I'm suspecting that this is not enough bandwidth. I'm doubting that it is latency alone that gives me all my trouble. I can handle high latency and low volume traffic, I can handle low latency and high volume traffic, but not the both together. That's a DC fest for me.
Only have personal experience I'm afraid...
Before I got my 8meg, I was using a 3mb HSDPA mobile broadband USB dongle, which came with a bandwidth monitor and it seemed that, in BG's, WoW generally used 5kBytes/s up
and
down and in Dalaran it was a little more down and the up depended completely on what I was doing, obviously... My ping was dependent on how much bandwidth was being used at the time but generally had 150ms doing 'normal' stuff and up to 500ms in Dalaran.
A 640 kbit/s line is, roughly translated, a 80kByte/s line so, in theory and depending on what else you're doing on your internet at that particular time, you shouldn't have any problems with lag in BGs or similar. Though, remember protocol headers/footers and that you probably won't be able to obtain your full connection speed any way..
However, as Obvious said, Wireless networks can be a &*!@# for online gaming and port forwarding can sometimes play up if your particular router's firewall is on...

If your playing wow via a wireless card/router, where either one is a g-based 54mbps, your actually usable bandwidth is only around 24mbps, due to overhead from the data header/footer. Adding known system resource hogs and poorly designed data bottlenecks, such as McCrappy and Bloatsaton, will futher increase the latentcy of 2 computers communicating over a distance.
Upgrade to an n-based (300mpbs) router and wireless card.

I'm having issues with people complaining that Ventrilo is lagging their game. I am a raidleader / co-raidleader and it happens quite often that people from the raid refuse to use Ventrilo as they claim it lags their game.
What I am wondering since I am no techno-wizard, is if anyone can tell me a way to minimize / diminish the lag Ventrilo occurs, or if Ventrilo does not drain on the Network at all. Since I do sometimes wonder if this is a simple excuse to avoid using Ventrilo.
Any advice much appreciated.

It's low on resources. You're a lot more likely to get lag from add-ons and other background programs before Vent gives you a lot of trouble. I know it isn't true for everyone, but the majority of people should be able to run Vent and WoW just fine without issue.

If you're running your own Ventrillo server, or even if you're paying for a service, you can adjust the quality of the audio transmitted; higher quality = more bandwidth used = more likely to lag.
That's is the only perceivable way you could lag from using Ventrilo... The only other factors being that the lagged player has crappy internet or loads of people are talking at once and it's spamming them.
tl;dr
It can lag you but it's unlikely it will.
