I normally start up with a 3 cp SnD and then a 5 cp rupture
After that I am doing 5 cp SnD and 4 cp Ruptures.
Why change from a 5 cp rupture to a 4 cp..?
I vary around 4/5s 5r 4/5e
Also link you armory, so we can check for ourselves that you are indeed correctly itemized

Other then swapping one red gem for an exp gem, one orange gem for an exp/hit gem and replacing the gem in your wrists for an orange ap/hit gem, i see no problem in your gear, talents nor glyphs.
So it must be you who is lacking something.
Ill give you the starting basics of combat.
Dont stealth, run in and start with one sinister strike and use that one cp for a snd.
Afterwards blow all your energy and use tricks, blow that gained energy too.
Quickly use killing spree, afterwards use a 5cp snd.
Either you can immediatly or you gotta ss one or two times. (Most importantly dont let snd drop)
Now blow blade flurry + adrenaline rush.
You will have enough energy for a 5cp rupture, a 5cp eviscerate (perhaps 2) and enough time to gain an other 5cp snd.
Cast snd and go for an other 5cp rupture, by now your rupture will have faded.
With the 5cp rupture used, you will have to squeeze in the remaining time an eviscerate.
This may be a 3cp or even a 5cp eviscerate. I am usually able to pull off 4/5cp eviscerate.
I cant give you a solid rotation, like in vanilla, now with the sinister strike glyph,
but this should give you a basic idea on what you should be doing.
Dont let snd drop and use tricks on cooldown.
Killing spree also too, but be sure to be energy starved first.
Dont be capping energy during killing spree, spam sinister strike or a finisher during killing spree, when you are reaching the energy cap. So that u use it immediatly after it.
A trick i use is: When snd is 4-5 secs remaining, then i wait a bit to gain energy (never cap) and use it at the last second. Using this instead of overclipping snd everytime, might save you a few dps.
Dont forget, spreadsheets dont accumulate human error.
They calculate your dps as if a robot would be playing.
If you got anymore questions, feel free to ask.

Hell, i don't even use rupture in my combat PvE and pull upwards of 7.5k+
Just SnD and eviscerate.
Different specc and the
rupture specc is mostly considered to be better.
Info about both speccs
